電顕写真 細胞内小器官(オルガネラ)の機能発現は各オルガネラに固有に存在している蛋白質に依存している。我々はリソソームへの蛋白質の選別・輸送機構およびその機能を明らかにすることを目的とした研究を行っている。リソソームは約50年前にC. de Duveによって発見されたオルガネラであり、現在では細胞内高分子分解の主要な場であるという位置付けが確立している。
一般的なリソソームの機能としては、細胞内外の生体高分子(オルガネラ・蛋白質・脂質・糖質など)の分解や、細胞死の際に自己消化に関わる消化酵素の供給などが知られている。さらに特殊化した細胞においてリソソームは、メラノソーム(メラニンの生成・蓄積)や消化酵素分泌顆粒として機能している。

蛍光写真(JCS) LGP85(赤色蛍光)の過剰発現により細胞内にコレステロール(青色蛍光)の蓄積した巨大空砲が形成される。Accumulation of cholesterol(blue) in LGP85 induced large vacuolar compartments(red). リソソーム機能の中核をなすのは、その内部に70種類以上存在する酸性加水分解酵素であり、これらリソソーム酵素の欠損は内部に未消化物が蓄積する「リソソーム蓄積症」と呼ばれる病変を招く。また、リソソームを形成する膜蛋白質の輸送異常や遺伝的欠陥でもリソソームの機能低下は認められ、特に筋肉や免疫系・血液凝固系の組織や細胞の機能不全による疾患が多くみられる。さらに、プリオン病、パーキンソン病、ハンチントン病などの神経性疾患の中には、蛋白質分解経路の破綻による異常蛋白質蓄積が原因となっているものが多数見つかっており、リソソーム系蛋白質分解研究の重要性を表わしている。

研究内容
  • リソソーム膜蛋白質の機能解析
    リソソーム膜蛋白質の機能を解明する目的でノックアウトマウスの作成や培養細胞による遺伝子発現を行う。最近我々はLGP85の過剰発現が巨大空胞形成を引き起こし(右上写真)、LGP85がリソソームの形態や機能を制御していることを報告した。
  • リソソームへの小胞輸送に関する研究
    リソソームへの物質輸送は小胞輸送によりエンドソームを経由して行われている。このような小胞輸送を制御するさまざまな分子の役割について研究を行っている。
  • 細胞死におけるリソソームの関与
    リソソームは「細胞の自殺袋」とも呼ばれ、細胞死において重要な役割を果たしている。細胞死におけるリソソームの役割を明らかにすることは癌などの病態との関連においても重要である。
  • メラノソーム形成機構に関する研究(美白の科学)
    メラノサイトにおいてメラノソームは「特殊リソソーム」であり、その形成は皮膚や頭髪の色素形成に重要な役割を果たしている。このようなメラノソームの形成機構を明らかにする。
Research

This laboratory focuses on lysosomes because they exhibit a number of important basic functions (digesting proteins, lipids, carbohydrates and organelles and supplying acid hydrolases for programmed cell death) as wel as having a highly specialized organization and functions in specialized cells (melanosomes in melanocytes, lytic granules in lymphocytes). We want to understand the molecular basis of lysosomal membrane proteins and how they contribute to cell physiology. Our initial approach was to study the function of specific lysosomal membrane proteins. We have prepared and used knockout mice to understand their physiological significance and found that LAMP2 function plays a role in a number of human diseases. We subsequently showed that LGP85 responds to membrane traffic to lysosomes via the cell expression system. An important goal is to identify the protein machinery that regulates membrane traffic to lysosomes. Using specific probes and materials (antibodies, ligands and cDNAs), we have been studying the molecular mechanism of membrane traffic to lysosomes and successfully identified several molecules which regulate membrane traffic to lysosomes. Our research has implications for some neurodegenerative diseases, since lysosome dysfunction is directly linked to many human diseases. Lysosomal biogenesis has relevance to virus budding, and thus our research also has many potential implications for viral pathogenesis.
